Q: Instead of specifying SETUP and HOLD time for data and clock in digital system, can we just specify a SETUP time for '1' and a SETUP time for '0'?

No. Setup and hold times do not relate to the level of the
the logic signal.
Setup time is how long a logic level must remain stable
prior to the event in question and hold time is how long
it must remain stable after the event to ensure that the
level will be read correctly.
